Quanterix flow cells are&nbsp;<a href=\"https:\/\/www.stratec.com\/references\/details\/Quanterix-7ea9e8fb-42c4a01a\">manufactured by ex-Sony DVD processing unit Stratec<\/a>. Stratec also manufacture&nbsp;<a href=\"https:\/\/www.stratec.com\/references\/details\/UnchainedLabs\">flow cells for the Lunatic<\/a>, a UV-Vis spectrophotometer made by Unchained Labs. And the name alone was enough to make me want to investigate further. Unchained was recently acquired for&nbsp;<a href=\"https:\/\/www.carlyle.com\/media-room\/news-release-archive\/carlyle-group-acquire-life-sciences-tools-company-unchained-labs\">$435 million<\/a>&nbsp;by Carlyle. The company expects to generate $75M in 2021 and has 170 employees.<\/p>\n\n\n\n<p>Unchained have a large portfolio of instruments. None of which I\u2019ve ever heard of, it\u2019s fascinating to me that companies like this can find relatively healthy niches while I assume having a relatively small market share.<\/p>\n\n\n\n<p>Unchained has a storied history, having acquired Trinean, Freeslate, and AVIA Biosystems. They have a portfolio of products covering UV-Vis, scattering, Raman based particle identification, and lab automation. I get the general impression that their focus is on drug development, which might explain why I\u2019ve not heard of them\u2026<\/p>\n\n\n\n<p>In this post, I wanted to delve into \u201cThe Lunatic\u201d a little. This is a UV-Vis spectrophotometer which they position against the Nanodrop.<\/p>\n\n\n\n<h1 class=\"wp-block-heading\">A Brief History of UV-Vis for Nucleic Acid Quantification<\/h1>\n\n\n\n<figure class=\"wp-block-embed is-type-rich is-provider-embed-handler wp-block-embed-embed-handler wp-embed-aspect-16-9 wp-has-aspect-ratio\"><div class=\"wp-block-embed__wrapper\">\n<span class=\"embed-youtube\" style=\"text-align:center; display: block;\"><iframe loading=\"lazy\" class=\"youtube-player\" width=\"640\" height=\"360\" src=\"https:\/\/www.youtube.com\/embed\/TGQplPeArz8?start=1s&#038;rel=1&#038;autoplay=0&#038;showinfo=1&#038;enablejsapi=0&#038;version=3&#038;showsearch=0&#038;iv_load_policy=1&#038;fs=1&#038;hl=en-US&#038;autohide=2&#038;wmode=transparent\" allowfullscreen=\"true\" style=\"border:0;\" sandbox=\"allow-scripts allow-same-origin allow-popups allow-presentation allow-popups-to-escape-sandbox\"><\/iframe><\/span>\n<\/div><\/figure>\n\n\n\n<p>The Lunatic is a UV-Vis spectrophotometer and appears to be marketed against the Nanodrop as a tool for detecting impurities in nucleic acids. The basic UV-Vis method looks at nucleic acid absorbance at 260nm and compares this to compares this to the absorbance from proteins and other contaminates peaking at 280nm.<\/p>\n\n\n\n<p>260nm is obviously way down in the UV range which means you need a UV capable spectrophotometer. This implies a UV light source, and UV compatible optics (fused silica) making the instrument more complex, and expensive.<\/p>\n\n\n\n<p>Typically these A260\/A280 measurements would be taken using a UV-Vis spectrophotometer such as the Beckman DU530 (which I\u2019ve been&nbsp;<a href=\"https:\/\/41j.com\/blog\/2021\/11\/beckman-du530-photospectrometer-notes\/\">tearing apart on my blog<\/a>). These instruments use light sources covering the visible and UV range. A grating splits this out and a slit is used to select a small range of wavelengths. The grating therefore has to physically move so as to direct a single part of the spectrum toward the slit.<\/p>\n\n\n\n<p>The instrument will scan across wavelength passing the monochromatic light through the sample and measuring absorbance. The photodiode then registers the amount of light absorbed one wavelength at a time. These instruments have a few issues, scans can take more than a minute, and you generally need at least 100ul of material.<\/p>\n\n\n\n<p>The Nanodrop provides a solution to these issues, requiring only 1ul of material, and giving a readout in a few seconds. It does this by using a different architecture:<\/p>\n\n\n\n<div class=\"wp-block-image\"><figure class=\"aligncenter size-full is-resized\"><img loading=\"lazy\" decoding=\"async\" src=\"https:\/\/41j.com\/blog\/wp-content\/uploads\/2021\/11\/luna2.png\" alt=\"\" class=\"wp-image-6400\" width=\"676\" height=\"231\" srcset=\"https:\/\/41j.com\/blog\/wp-content\/uploads\/2021\/11\/luna2.png 901w, https:\/\/41j.com\/blog\/wp-content\/uploads\/2021\/11\/luna2-300x103.png 300w, https:\/\/41j.com\/blog\/wp-content\/uploads\/2021\/11\/luna2-150x51.png 150w, https:\/\/41j.com\/blog\/wp-content\/uploads\/2021\/11\/luna2-768x263.png 768w\" sizes=\"auto, (max-width: 676px) 100vw, 676px\" \/><\/figure><\/div>\n\n\n\n<p>Essentially spread spectrum light is sent through the sample, some is absorbed and the remaining light comes out the other side. Only then is a grating used to split the light and the absorbance spectrum measured. The spectra is registered on a linear image sensor (CCD, or potentially a CMOS sensor in newer instruments). No moving parts are required, and measurement is near instantaneous.<\/p>\n\n\n\n<p>Rather than using a cuvette (sample container) on the Nanodrop the sample is directly sandwiched between two fiber optic cables, which allows for very low sample volume:<\/p>\n\n\n\n<div class=\"wp-block-image\"><figure class=\"aligncenter size-full\"><img loading=\"lazy\" decoding=\"async\" width=\"457\" height=\"378\" src=\"https:\/\/41j.com\/blog\/wp-content\/uploads\/2021\/11\/luna4.jpg\" alt=\"\" class=\"wp-image-6401\" srcset=\"https:\/\/41j.com\/blog\/wp-content\/uploads\/2021\/11\/luna4.jpg 457w, https:\/\/41j.com\/blog\/wp-content\/uploads\/2021\/11\/luna4-300x248.jpg 300w, https:\/\/41j.com\/blog\/wp-content\/uploads\/2021\/11\/luna4-150x124.jpg 150w\" sizes=\"auto, (max-width: 457px) 100vw, 457px\" \/><\/figure><\/div>\n\n\n\n<p>For general purpose UV-Vis applications, this approach likely has a number of disadvantages, but for nucleic acid quantification it appears to work just fine.<\/p>\n\n\n\n<p>The Nanodrop doesn\u2019t require any consumables, and from what I understand is a pretty robust instrument, rarely requiring servicing.&nbsp;<\/p>\n\n\n\n<h1 class=\"wp-block-heading\">The Lunatic<\/h1>\n\n\n\n<p>The Lunatic sets itself up as an alternative to the Nanodrop. This is a microfluidic chip based system which performs a similar function to the Nanodrop, and similarly is marketed toward nucleic acid quantification:<\/p>\n\n\n\n<figure class=\"wp-block-embed is-type-video is-provider-youtube wp-block-embed-youtube wp-embed-aspect-16-9 wp-has-aspect-ratio\"><div class=\"wp-block-embed__wrapper\">\n<span class=\"embed-youtube\" style=\"text-align:center; display: block;\"><iframe loading=\"lazy\" class=\"youtube-player\" width=\"640\" height=\"360\" src=\"https:\/\/www.youtube.com\/embed\/OTBWDwtN7To?version=3&#038;rel=1&#038;showsearch=0&#038;showinfo=1&#038;iv_load_policy=1&#038;fs=1&#038;hl=en-US&#038;autohide=2&#038;wmode=transparent\" allowfullscreen=\"true\" style=\"border:0;\" sandbox=\"allow-scripts allow-same-origin allow-popups allow-presentation allow-popups-to-escape-sandbox\"><\/iframe><\/span>\n<\/div><\/figure>\n\n\n\n<p>There are different chips available for the Lunatic, but they all essentially draw the solution into a detection region. As far as I can tell there\u2019s no active fluidics in this system, it\u2019s all driven by capillary action:<\/p>\n\n\n\n<div class=\"wp-block-image\"><figure class=\"aligncenter size-medium\"><img loading=\"lazy\" decoding=\"async\" width=\"300\" height=\"115\" src=\"https:\/\/41j.com\/blog\/wp-content\/uploads\/2021\/11\/luna5-300x115.jpg\" alt=\"\" class=\"wp-image-6402\" srcset=\"https:\/\/41j.com\/blog\/wp-content\/uploads\/2021\/11\/luna5-300x115.jpg 300w, https:\/\/41j.com\/blog\/wp-content\/uploads\/2021\/11\/luna5-1024x394.jpg 1024w, https:\/\/41j.com\/blog\/wp-content\/uploads\/2021\/11\/luna5-150x58.jpg 150w, https:\/\/41j.com\/blog\/wp-content\/uploads\/2021\/11\/luna5-768x295.jpg 768w, https:\/\/41j.com\/blog\/wp-content\/uploads\/2021\/11\/luna5.jpg 1072w\" sizes=\"auto, (max-width: 300px) 100vw, 300px\" \/><\/figure><\/div>\n\n\n\n<p>Unchained labs suggest that the Lunatic has similar sensitivity and reproducibility to the NanoDrop. But that the Nanodrop shows significant variation between instruments:<\/p>\n\n\n\n<div class=\"wp-block-image\"><figure class=\"aligncenter size-large is-resized\"><img loading=\"lazy\" decoding=\"async\" src=\"https:\/\/41j.com\/blog\/wp-content\/uploads\/2021\/11\/luna6-1024x440.png\" alt=\"\" class=\"wp-image-6403\" width=\"512\" height=\"220\" srcset=\"https:\/\/41j.com\/blog\/wp-content\/uploads\/2021\/11\/luna6-1024x440.png 1024w, https:\/\/41j.com\/blog\/wp-content\/uploads\/2021\/11\/luna6-300x129.png 300w, https:\/\/41j.com\/blog\/wp-content\/uploads\/2021\/11\/luna6-150x64.png 150w, https:\/\/41j.com\/blog\/wp-content\/uploads\/2021\/11\/luna6-768x330.png 768w, https:\/\/41j.com\/blog\/wp-content\/uploads\/2021\/11\/luna6.png 1456w\" sizes=\"auto, (max-width: 512px) 100vw, 512px\" \/><\/figure><\/div>\n\n\n\n<p>In some ways this feels like a calibration issue, in that the Nanodrops are showing the necessary reproducibility, but that they are not typically calibrated to the same extent as the Lunatic. It\u2019s also not clear to me that the Nanodrop is the right instrument to compare the Lunatic with, perhaps&nbsp;<a href=\"https:\/\/www.denovix.com\/blog\/nanodrop-competitors-microvolume-analysis\/\">Denovix who position themselves as a higher sensitivity alternative<\/a>&nbsp;might make a more interesting comparison. The Lunatic does appear to have a higher dynamic range than the Nanodrop, and this maybe useful in some scenarios.<\/p>\n\n\n\n<p>The major disadvantage of the Lunatic of course is that you need to buy chips to use it. From what I can tell, this works out at&nbsp;<a href=\"https:\/\/www.fishersci.com\/shop\/products\/dropplatestrips-s-16-box-30\/501125947\">about $4 per sample<\/a>. The chip likely as some advantage in terms of ease of use and clean up, but overall I find it hard to see a strong advantage over the Nanodrop and other platforms.<\/p>\n\n\n\n<p>But with this instrument being marketed toward drug discovery, perhaps even a small edge over the Nanodrop coupled with a healthy portfolio of instruments, is enough.<\/p>\n","protected":false},"excerpt":{"rendered":"<p>Unchained Labs I originally came across the Lunatic when investigating&nbsp;Quanterix\u2019s flow cells.
